iTL specializes in the creation of custom-made mouse knockout (conventional, conditional) and knockin (point mutation,human cDNA replacement,reporter gene) models. In
addition, many investigators create their own targeting constructs and
have iTL perform the remaining steps, i.e. from electroporation onward.
ITL is also capable of creating just targeting vectors, for
investigators who want to use another source for the rest of the
process.
In 2005, iTL introduced their Hybrid 129SvEv x C57BL/6 line. This
line exhibits a significantly higher rate of homologous recombination
in the production of mouse knockout and knockin models. The Hybrid
129SvEv x C57BL/6 has shown an average 200% increase in the frequency
of positive clones per targeting event than other cell lines used at
iTL. These characteristics have resulted in reducing the time involved
for electroporation and screening tasks, enabling delivery of mouse
models with guaranteed germline transmission to be achieved in as
little as 7 months. Another benefit iTL offers with this cell line is
the ability to provide scientists with knockout or knockin mouse models
at lower cost.
The Hybrid 129SvEv x C57BL/6 line is the third ES cell line developed
by iTL and is designed to incorporate the strengths of its 129SvEv and
C57BL/6 ES lines. The 129SvEv line exhibits a consistent rate of
homologous recombination for efficient germline transmission while
enabling the ability to analyze resulting chimeras by coat color. The
C57BL/6 line is useful for its physiological relevance to certain
genetically-linked health conditions, its many parallels between mouse
and human genes, the accuracy of published databases for C57BL/6 genes,
and the ability to obtain a mouse on a pure background.
