Knock-In Mouse Models
Knock-in mice continue to be powerful research models. The ability to insert an expanding variety of functional genetic components has broadened the spectrum of research programs utilizing these custom research tools.
iTL has particular strength and experience in these manipulations:
- Humanization. Insertion of human genomic or cDNA to replace an endogenous mouse gene
- Murine. Insertion of murine cDNA or replacement of targeted murine gene(s) with other murine gene
- In-frame fusion. A reporter gene is fused to the first coding exon
- Targeted overexpression. A transgene is inserted into a well characterized locus
- Point mutation(s)
- IRES, 2A peptide, or other tags. Strategies to facilitate the coexpression of a cDNA/reporter with an endogenous gene
- Reporter gene insertion. Reporter genes such as GFP, lacZ, mCherry, tdTomato, YFP and DsRed can be inserted either to replace an endogenous gene or to be fused to the endogenous gene for coexpression. iTL has also created a number of conditional knock-out models with reporter genes for our customers.
- Combination of Above. We have the expertise to combine multiple manipulations in a single model to provide you a fully custom model specialized to your research
These insertions and manipulations yield custom mouse models possessing specialized alterations in gene function tailored to your research goals.
