Conditional&Coventional Knockout/Knockin Mouse

The creation of knockout and knockin mice requires a wide range of expertise. iTL has had a high success rate using our proprietary ES cell lines.

iTL creates Conventional and Conditional knockout mice, Knockin mice, and Conventional/Conditional knockout mice with a reporter gene. In addition, many investigators create their own targeting constructs and have iTL perform the remaining steps, i.e. from electroporation onward. ITL is also capable of creating just targeting vectors, for investigators who want to use another source for the rest of the process.

    Conventional Knockout Mice DiagramConventional Knockout Mice: A mouse model in which a targeted gene has been functionally eliminated by inserting the selection cassette or a reporter gene such as GFP or LacZ into the locus of the gene. These mice can be used to study the unique function of a specific gene within the animal's genome.


    Conditional Knockout Mice DiagramConditional Knockout Mice: A mouse model that utilizes the cre-lox system (site specific DNA recombination through the interaction of the Cre recombinase enzyme and lox-P sites in the DNA strand for the deletion of a gene in targeted tissues).



    Knockin Mice DiagramKnockin Mice: A mouse model in which the targeted gene sequence has been subtly altered to modify its function. iTL can perform the following knockin manipulations:
    - Introduction of specific point mutations
    - Humanization by insertion of human cDNA to replace the endogenous mouse gene
    - Targeted Overexpression using Rosa26 in which a transgene is inserted into the independent Rosa 26 locus
    - Fusing a reporter gene to the first coding exon (In-frame Fusion)


    Conventional/Conditional Knockout Mice with Reporter GeneConventional/Conditional Knockout Mice with Reporter Gene: A mouse model that contains a reporter gene such as the beta-galactosidase reporter (Lac-Z), Luciferase or other fluorescent reporters, which serves as a marker for target gene expression. Genes can either be replaced by the reporter gene so only the reporter is expressed under the endogenous mouse promoter, or the reporter can be fused to an endogenous gene of interest so both the reporter and the mouse gene are expressed under the endogenous promoter.


    Partial Services: If investigators choose to create their own conventional, conditional, or knockin targeting vector(s), they can contract iTL to perform the services of electroporation, tissue culture, microinjection, breeding of chimeras, and germline confirmation. In addition, iTL can create the targeting vector, leaving investigators with the option of using their core facility for the rest of the process.
