Why Model Adora2a

Adora2a is the mouse ortholog of human ADORA2A and encodes the adenosine A2A receptor, a class A G-protein-coupled receptor. A2A-receptor signaling has been associated with regulation of immune-cell activity and with neurological functions in the basal ganglia.

A2A-receptor antagonists are used in Parkinson’s disease and are under study in cancer immunotherapy, where adenosine signaling can dampen immune responses. Adora2a-deficient mice are viable and show altered responses to adenosine and changes in motor and inflammatory phenotypes.

Model Applications

Humanized ADORA2A models, in which the human receptor replaces the mouse counterpart, have been used to evaluate A2A-receptor-targeted agents in defined experimental settings.
