Why Model C5

C5 is the mouse ortholog of human C5 and encodes complement component C5, a protein of the terminal complement pathway. C5 is cleaved to generate the anaphylatoxin C5a and the fragment C5b, which initiates assembly of the membrane attack complex, and has been associated with inflammation and cell lysis.

The terminal complement pathway, including C5, is implicated in paroxysmal nocturnal hemoglobinuria, atypical hemolytic uremic syndrome, and other conditions, and C5-directed antibodies are in clinical use. Mouse strains deficient in C5 activity show altered complement-dependent responses and susceptibility to certain challenges.

Model Applications

Humanized C5 models, in which the human protein replaces the mouse counterpart, have been used to evaluate C5-targeted therapeutics in defined experimental settings.

C5 models have been used to study terminal complement activity.
