Why Model Cd40

Cd40 is the mouse ortholog of human CD40 and encodes CD40, a TNF-receptor-superfamily member expressed on antigen-presenting cells and other cells. CD40 binds CD40 ligand (CD154) and has been associated with activation of antigen-presenting cells and support of T-cell and B-cell responses.

CD40 agonists are under study in cancer immunotherapy, and the CD40-CD40L axis is also implicated in inflammatory and vascular disease. Cd40-deficient mice are viable and show impaired antibody class switching and germinal-center responses.

Model Applications

Humanized CD40 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate agonistic CD40 antibodies in immunocompetent mice, sometimes combined with CD40L humanization.
