Why Model Cd47

Cd47 is the mouse ortholog of human CD47 and encodes CD47 (integrin-associated protein), a widely expressed cell-surface protein. CD47 binds the myeloid receptor SIRPα and has been associated with a signal that limits macrophage phagocytosis, sometimes described as a ‘don’t-eat-me’ signal.

CD47 overexpression on tumor cells has been associated with evasion of macrophage clearance in some contexts, and CD47-SIRPα-targeted agents have been evaluated in cancer; reported clinical results have varied. Cd47-deficient mice are viable and show altered clearance of circulating cells.

Model Applications

Humanized CD47 models, often combined with SIRPα humanization, have been used to evaluate CD47-SIRPα-targeted therapeutics and to support engraftment of human cells.

Cd47 knockout mice have been used to study macrophage regulation in defined experimental settings.
