Why Model CXCR4

Cxcr4 is the mouse ortholog of human CXCR4 and encodes CXCR4 (CD184), a class A G-protein-coupled chemokine receptor that binds the chemokine CXCL12 (SDF-1). CXCR4 has been associated with hematopoietic cell trafficking, retention of stem cells in bone marrow, and immune-cell migration; it also serves as a co-receptor for certain strains of HIV.

CXCR4 is implicated in cancer metastasis and in WHIM syndrome, and CXCR4-directed agents are used for stem-cell mobilization and are under study in cancer. Homozygous Cxcr4-null mice show embryonic lethality with defects in hematopoiesis and vascular and neural development.

Model Applications

Humanized CXCR4 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate CXCR4-directed agents in defined experimental settings.

Conditional Cxcr4 models have been used to study cell trafficking and hematopoiesis.
