Why Model DLL4
Dll4 is the mouse ortholog of human DLL4 and encodes Delta-like 4, a membrane-bound ligand of the Notch pathway expressed in arterial and tumor endothelium. DLL4-Notch signaling has been associated with regulation of blood-vessel branching and development.
DLL4 has been associated with tumor angiogenesis, and DLL4-directed agents have been evaluated in cancer. Heterozygous loss of Dll4 causes vascular defects and embryonic lethality in mice, consistent with a dose-sensitive role in vascular development.
Model Applications
Humanized DLL4 models, in which the human protein replaces the mouse counterpart, have been used to evaluate DLL4-directed agents in defined experimental settings.
