Why Model Gpc3
Gpc3 is the mouse ortholog of human GPC3 and encodes glypican-3, a GPI-anchored heparan sulfate proteoglycan. Glypican-3 has been associated with regulation of cell growth and signaling during development, and loss-of-function variants are linked to an overgrowth syndrome.
Glypican-3 is expressed on most hepatocellular carcinomas with limited expression in normal adult liver, and glypican-3-directed agents, including CAR T cells and antibodies, have been evaluated; clinical results have varied. Gpc3-deficient mice show developmental overgrowth features.
Model Applications
Humanized GPC3 models, in which the human protein replaces the mouse counterpart, have been used to evaluate glypican-3-targeted therapeutics in defined experimental settings.
