Why Model LGR5

Lgr5 is the mouse ortholog of human LGR5 and encodes a leucine-rich-repeat G-protein-coupled receptor that binds R-spondins and potentiates Wnt signaling. LGR5 marks stem cells in the intestine and other tissues and has been associated with tissue renewal.

LGR5 has been associated with cancer-stem-cell populations and treatment resistance in colorectal and other cancers, and LGR5-directed approaches are under study. Lgr5 reporter and lineage-tracing mice are widely used in stem-cell research.

Model Applications

Humanized LGR5 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate LGR5-directed agents in defined experimental settings.

Lgr5 reporter mice are used to track and study stem cells.
