Why Model MPL

Mpl is the mouse ortholog of human MPL and encodes the thrombopoietin receptor (CD110, TPOR), a type I cytokine receptor on megakaryocytes and hematopoietic progenitors. MPL binds thrombopoietin and has been associated with platelet production and hematopoietic stem-cell maintenance.

MPL mutations are associated with myeloproliferative neoplasms and inherited thrombocytopenias, and thrombopoietin-receptor agonists are used for immune thrombocytopenia and other conditions. Mpl-deficient mice show reduced platelets and megakaryocytes.

Model Applications

Humanized MPL models, in which the human receptor replaces the mouse counterpart, have been used to evaluate thrombopoietin-receptor-directed agents in defined experimental settings.
