Why Model Tnfrsf17

Tnfrsf17 is the mouse ortholog of human TNFRSF17 and encodes BCMA (B-cell maturation antigen, CD269), a TNF-receptor-superfamily member expressed mainly on plasma cells and late-stage B cells. BCMA binds the ligands BAFF and APRIL and has been associated with plasma-cell survival and antibody responses.

BCMA is expressed on malignant plasma cells in multiple myeloma, and BCMA-directed CAR T cells and bispecific antibodies are used in relapsed or refractory disease; responses vary and relapse can occur. Tnfrsf17-deficient mice are viable with reduced numbers of long-lived plasma cells.

Model Applications

Humanized TNFRSF17 (BCMA) models, in which the human protein replaces the mouse counterpart, have been used to evaluate BCMA-directed CAR T cells and bispecific antibodies in immunocompetent mice, including in combination settings.

Tnfrsf17 models have been used to study plasma-cell biology in defined experimental settings.
