Frequently asked questions

Consider detection method (fluorescence for live imaging, LacZ for fixed tissue), expression level (high-sensitivity reporters like LacZ for low expression), resolution needs (single-cell vs tissue-level), stability requirements (long-term expression for lineage tracing), and downstream applications (cell sorting, imaging, histology).

Fluorescent reporters (GFP, tdTomato, mCherry) enable live imaging, cell sorting, and real-time tracking but require specialized equipment. Enzymatic reporters (LacZ) provide excellent sensitivity for rare cells, work in fixed tissue, and don't require specialized imaging equipment but are destructive (tissue must be fixed and stained).

tdTomato and other stable fluorescent proteins are excellent for lineage tracing due to long-term stability and bright signal. LacZ can also be used for lineage tracing with whole-mount staining. Selection depends on whether you need live imaging (fluorescent) or fixed tissue analysis (LacZ).

Yes. Reporters can be made conditional using LoxP-stop-LoxP cassettes that prevent expression until Cre-mediated recombination removes the stop.
