Why Model DKK1
Dkk1 is the mouse ortholog of human DKK1 and encodes Dickkopf-1, a secreted antagonist of the Wnt/β-catenin pathway. DKK1 has been associated with regulation of Wnt signaling in development and bone.
DKK1 has been associated with bone loss in multiple myeloma and with roles in several cancers and immune regulation, and DKK1-directed agents are under study. Dkk1-deficient and mutant mice show skeletal and developmental phenotypes.
Model Applications
Humanized DKK1 models, in which the human protein replaces the mouse counterpart, have been used to evaluate DKK1-directed agents in defined experimental settings.
