Why model Dkk1
Dkk1, the mouse ortholog of human DKK1, encodes a secreted glycoprotein with two cysteine rich domains that antagonizes canonical Wnt signaling by binding the third and fourth beta-propeller modules of LRP5 and LRP6 with high affinity, blocking Wnt induced LRP6 signalosome assembly with Frizzled, and in some contexts cooperating with Kremen1 and Kremen2 to internalize the coreceptor. Germline null embryos die at birth lacking anterior head structures and with limb malformation because DKK1 is required for head organizer activity, so conditional deletion is necessary for adult bone and hematopoietic questions. Dkk2 and Dkk4 bind the same LRP sites, with DKK2 acting as agonist or antagonist depending on Kremen levels, so compound alleles matter.
