Why Model Flt3

Flt3 is the mouse ortholog of human FLT3 and encodes FLT3 (CD135), a class III receptor tyrosine kinase expressed on hematopoietic progenitor cells. Binding of FLT3 ligand promotes signaling that has been associated with proliferation and survival of early progenitors.

Activating FLT3 mutations, including internal tandem duplications, have been associated with acute myeloid leukemia, and FLT3 inhibitors are used in FLT3-mutated disease; resistance can emerge. Flt3-deficient mice are viable with reduced numbers of certain progenitor populations.

Model Applications

Humanized FLT3 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate FLT3-targeted agents.

Flt3 models have been used to study hematopoiesis and FLT3-driven leukemia in defined experimental settings.
