Why Model Lrrk2

Lrrk2 is the mouse ortholog of human LRRK2 and encodes a large protein containing both GTPase and kinase domains. LRRK2 has been associated with regulation of vesicle trafficking, lysosomal function, and other cellular processes.

LRRK2 mutations, including the common G2019S variant, are among the more frequent genetic contributors to Parkinson’s disease, and LRRK2 kinase inhibitors are under clinical investigation. Lrrk2-deficient mice are viable and show changes in lung and kidney and in lysosomal markers.

Model Applications

Humanized and mutant LRRK2 models, including knock-in of disease-associated variants, have been used to study LRRK2 biology and to evaluate LRRK2-targeted agents in defined experimental settings.

Lrrk2 knockout models have been used to study LRRK2 function.
