Why Model MADCAM1
Madcam1 is the mouse ortholog of human MADCAM1 and encodes mucosal addressin cell adhesion molecule-1 (MAdCAM-1), an immunoglobulin-superfamily adhesion molecule expressed on endothelium in the gut and associated lymphoid tissue. MAdCAM-1 binds the α4β7 integrin and has been associated with lymphocyte homing to the intestine.
The MAdCAM-1/α4β7 axis is a target in inflammatory bowel disease, and MAdCAM-1-directed agents have been evaluated. Madcam1-deficient mice show altered gut lymphocyte homing.
Model Applications
Humanized MADCAM1 models, in which the human protein replaces the mouse counterpart, have been used to evaluate MAdCAM-1-directed agents in defined experimental settings.
