Why Model MUC1

Muc1 is the mouse ortholog of human MUC1 and encodes mucin 1 (CD227), a transmembrane glycoprotein expressed on the apical surface of many epithelia. MUC1 has been associated with lubrication, barrier protection, and cell signaling.

MUC1 is overexpressed and aberrantly glycosylated on many carcinomas, and MUC1-directed antibodies, antibody–drug conjugates, vaccines, and cell therapies have been evaluated; clinical results have varied. Muc1-deficient mice are viable with increased susceptibility to certain infections.

Model Applications

Humanized MUC1 models, which express human MUC1 (including tumor-associated forms), have been used to study MUC1 biology and to evaluate MUC1-directed therapeutics in defined experimental settings.
