Why Model Nt5e
Nt5e is the mouse ortholog of human NT5E and encodes CD73, a GPI-anchored ecto-5’-nucleotidase that converts AMP to adenosine. CD73, together with CD39, has been associated with generation of extracellular adenosine and with regulation of immune responses.
The CD39-CD73-adenosine pathway has been associated with an immunosuppressive tumor microenvironment, and CD73-directed agents are under study in cancer. Nt5e-deficient mice are viable and show altered vascular permeability and immune responses.
Model Applications
Humanized NT5E (CD73) models, in which the human protein replaces the mouse counterpart, have been used to evaluate CD73-targeted agents in defined experimental settings.
