Why Model Tnfrsf9

Tnfrsf9 is the mouse ortholog of human TNFRSF9 and encodes 4-1BB (CD137), a costimulatory TNF-receptor-superfamily member expressed on activated T cells and other immune cells. Engagement of 4-1BB by its ligand has been associated with enhanced T-cell proliferation and survival.

4-1BB is a target of agonist antibodies and bispecific agents in cancer immunotherapy, and the 4-1BB signaling domain is used in some chimeric antigen receptors. Tnfrsf9-deficient mice are viable with altered T-cell responses in some models.

Model Applications

Humanized TNFRSF9 (4-1BB) models, in which the human receptor replaces the mouse counterpart, have been used to evaluate 4-1BB agonists and bispecific agents in immunocompetent mice.
