Cd19 Conditional Knockout mouse models

Cd19, ortholog of human CD19, encodes a B lineage immunoglobulin superfamily coreceptor with two extracellular Ig-like domains that partners with Cr2, Cd81 and Cd225 to lower the B cell receptor activation threshold, recruiting PI3K to phosphorylated cytoplasmic tyrosines and amplifying Btk and Akt signaling. Its dominant translational role is as the antigen for CD19-directed CAR T cells and for blinatumomab, and because those biologics recognize human epitopes that murine Cd19 does not present, an epitope-matched humanized CD19 knockin is the only credible platform for efficacy, B cell aplasia and cytokine release studies. A conventional null instead answers a developmental question, yielding reduced marginal zone and B1 populations with blunted humoral responses. Investigators should also note that Cd19-Cre is a knockin driver, so the Cre allele itself is a functional null and heterozygosity must be controlled. Pair alleles with flow cytometric B cell subset analysis, antigen-specific titers, and CAR persistence readouts.

Tissue restricted knockout of Cd19 keeps wild type function everywhere else. Breeding is often more robust, and the setup mirrors patient biology where mutations arise in a subset of cells.
