Cstb Knockout mouse models

Cstb is the mouse ortholog of human CSTB and encodes cystatin B (stefin B), a small monomeric cytosolic inhibitor of papain-family cysteine cathepsins that uses a wedge formed by its N-terminal glycine region and two hairpin loops to occlude the active sites of cathepsins B, L, S, and H, guarding against leaked lysosomal proteolysis. Loss produces oxidative stress, lysosomal instability, and cerebellar granule cell apoptosis, and mouse nulls develop progressive myoclonus, ataxia, and microglial activation reproducing human Unverricht-Lundborg disease, EPM1. Note a modeling constraint: most human EPM1 alleles are expansions of a dodecamer repeat in the CSTB promoter that reduce transcription, a mutation class that simple coding deletion cannot replicate, so promoter humanization or knockin of the repeat is required for expression-level fidelity. Related cystatins do not compensate in brain, so use the null for neurodegeneration endpoints and conditional alleles to test neuronal versus microglial origin.
