Why Model AGT
Agt is the mouse ortholog of human AGT and encodes angiotensinogen, a secreted precursor of the renin-angiotensin system produced mainly by the liver. Angiotensinogen is cleaved by renin to generate angiotensin I and has been associated with regulation of blood pressure and fluid balance.
The renin-angiotensin system is central to blood-pressure regulation, and angiotensinogen is targeted by RNA-interference approaches under study for hypertension. Agt-deficient mice show low blood pressure and kidney abnormalities.
Model Applications
Humanized AGT models, in which the human protein replaces the mouse counterpart, have been used to evaluate angiotensinogen-directed agents in defined experimental settings.
