Why Model REN

REN encodes renin, an aspartyl protease secreted mainly by the kidney; mouse strains carry one or two renin genes (Ren1, Ren2). Renin cleaves angiotensinogen to generate angiotensin I and has been associated with the rate-limiting step of the renin-angiotensin system.

The renin-angiotensin system is central to blood-pressure and fluid regulation, and renin and its pathway are targeted by several antihypertensive therapies. Mice with altered renin genes show changes in blood pressure and kidney physiology.

Model Applications

Humanized REN models, in which the human gene replaces the mouse counterpart, have been used to study renin biology and to evaluate renin-system-directed agents in defined experimental settings.
