Why Model ASGR1

Asgr1 is the mouse ortholog of human ASGR1 and encodes the major subunit of the asialoglycoprotein receptor, a C-type lectin on hepatocytes that binds galactose- and N-acetylgalactosamine-terminated glycoproteins and mediates their uptake. ASGR1 has been associated with clearance of desialylated glycoproteins.

ASGR1 is used for liver-targeted drug delivery (for example, GalNAc-conjugated therapeutics), and loss-of-function ASGR1 variants have been associated with lower non-HDL cholesterol and cardiovascular risk. Asgr1-deficient mice show altered glycoprotein clearance.

Model Applications

Humanized ASGR1 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate ASGR1-targeted delivery and agents in defined experimental settings.
