Why Model Cd274
Cd274 is the mouse ortholog of human CD274 and encodes PD-L1 (B7-H1), a cell-surface ligand expressed on various immune, epithelial, and tumor cells. PD-L1 binds the inhibitory receptor PD-1 and has been associated with downregulation of T-cell responses.
PD-L1 expression on tumor cells has been associated with immune evasion in some contexts, and antibodies targeting PD-L1 are approved for several cancers; responses depend on tumor type and other factors. Cd274-deficient mice are viable and show altered T-cell responses in disease models.
Model Applications
Humanized CD274 models, in which the human ligand replaces the mouse counterpart, have been used to evaluate anti-PD-L1 antibody therapeutics in immunocompetent mice, including in combination with other checkpoint-targeted agents.
Cd274 knockout mice have been used to study the ligand’s role in immune regulation.
