Why Model CFB
Cfb is the mouse ortholog of human CFB and encodes complement factor B, a serine protease zymogen of the alternative complement pathway. Factor B contributes to formation of the alternative-pathway C3 convertase and has been associated with amplification of complement activation.
The alternative pathway, including factor B, is implicated in several complement-mediated diseases, and factor B inhibitors are in clinical use and development. Cfb-deficient mice show impaired alternative-pathway activity.
Model Applications
Humanized CFB models, in which the human protein replaces the mouse counterpart, have been used to evaluate factor B-directed agents in defined experimental settings.
