Why Model CFH
Cfh is the mouse ortholog of human CFH and encodes complement factor H, a soluble regulator of the alternative complement pathway. Factor H has been associated with restraining complement activation on host surfaces.
CFH variants are associated with age-related macular degeneration and with complement-mediated kidney diseases, and complement-targeted therapies are relevant to these conditions. Cfh-deficient mice develop complement dysregulation and kidney disease resembling C3 glomerulopathy.
Model Applications
Humanized CFH models, in which the human protein replaces the mouse counterpart, have been used to study factor H regulation and to evaluate complement-targeted approaches in defined experimental settings.
