Why Model Dpp4
Dpp4 is the mouse ortholog of human DPP4 and encodes dipeptidyl peptidase-4 (CD26), a cell-surface and soluble serine exopeptidase. DPP-4 cleaves several peptides, including the incretin hormones GLP-1 and GIP, and has been associated with regulation of their activity.
DPP-4 inhibitors are used in type 2 diabetes, and DPP-4 has additional roles in immune regulation; in some species DPP-4 serves as a receptor for certain coronaviruses. Dpp4-deficient mice are viable and show altered glucose tolerance and incretin levels.
Model Applications
Humanized DPP4 models, in which the human protein replaces the mouse counterpart, have been used to study DPP-4 biology, DPP-4-targeted agents, and human DPP-4-dependent coronavirus infection in defined experimental settings.
