Why Model Glp1r

Glp1r is the mouse ortholog of human GLP1R and encodes the glucagon-like peptide-1 receptor, a class B G-protein-coupled receptor. GLP-1R signaling, primarily through Gs and cAMP, has been associated with glucose-dependent insulin secretion, slowed gastric emptying, and reduced food intake.

GLP-1 receptor agonists are used in type 2 diabetes and obesity, and the receptor is a target of several marketed and investigational agents. Glp1r-deficient mice are viable and show altered glucose tolerance and insulin secretion.

Model Applications

Humanized GLP1R models, in which the human receptor replaces the mouse counterpart, have been used to evaluate GLP-1-receptor-targeted peptides and small molecules in vivo.

Glp1r knockout and reporter models have been used to study incretin signaling and receptor distribution.
