Why Model Folr1
Folr1 is the mouse ortholog of human FOLR1 and encodes folate receptor alpha, a GPI-anchored receptor that binds folate with high affinity and mediates its cellular uptake. Folate receptor alpha has been associated with folate transport in selected epithelial tissues.
Folate receptor alpha is overexpressed on several tumors, including ovarian cancer, and folate-receptor-alpha-directed antibody–drug conjugates are used in selected patients. Folr1-deficient mice show developmental defects related to folate transport.
Model Applications
Humanized FOLR1 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate folate-receptor-alpha-targeted agents, including antibody–drug conjugates, in defined experimental settings.
