Why Model Msln

Msln is the mouse ortholog of human MSLN and encodes mesothelin, a GPI-anchored cell-surface glycoprotein normally expressed on mesothelial cells. Mesothelin has been associated with cell adhesion, and its normal physiological role is not fully defined.

Mesothelin is overexpressed on several solid tumors, including mesothelioma and ovarian and pancreatic cancers, and mesothelin-directed antibody–drug conjugates, CAR T cells, and antibodies have been evaluated; clinical results have varied. Msln-deficient mice are viable without an overt phenotype under standard conditions.

Model Applications

Humanized MSLN models, in which the human protein replaces the mouse counterpart, have been used to evaluate mesothelin-targeted therapeutics, including antibody–drug conjugates and cell therapies, in defined experimental settings.
