Why Model Lag3

Lag3 is the mouse ortholog of human LAG3 and encodes LAG-3 (CD223), an inhibitory receptor expressed on activated T cells and other lymphocytes. LAG-3 binds ligands including MHC class II and has been associated with negative regulation of T-cell responses.

LAG-3 is a target of cancer immunotherapy, and an anti-LAG-3 antibody is approved in combination with PD-1 blockade for melanoma. Lag3-deficient mice are viable and show altered T-cell regulation in some models.

Model Applications

Humanized LAG3 models, in which the human receptor replaces the mouse counterpart, have been used to evaluate anti-LAG-3 therapeutics, including in combination with PD-1-targeted agents, in immunocompetent mice.

Lag3 knockout mice have been used to study its role in T-cell regulation.
