Why Model Mapt

Mapt is the mouse ortholog of human MAPT and encodes tau, a microtubule-associated protein enriched in neurons. Tau has been associated with regulation of microtubule stability and axonal transport, and it is expressed as multiple isoforms.

Aggregated tau is a feature of Alzheimer’s disease and other tauopathies, and MAPT mutations are linked to frontotemporal dementia; tau-directed therapies are under study, though clinical results to date have been mixed. Humanized and mutant Mapt alleles have been developed to study tau biology.

Model Applications

Humanized MAPT models, which express human tau isoforms, have been used to study tau aggregation and to evaluate tau-targeted approaches in defined experimental settings.

Mapt knockout and mutant models have been used to study tau function.
