Acvr2b, mouse ortholog of human ACVR2B, encodes the type IIB activin receptor, the highest affinity receptor for myostatin and GDF11 and a principal node controlling skeletal muscle mass, axial patterning and left-right asymmetry. Ligand capture drives constitutive kinase activity that phosphorylates ALK4 or ALK5, mobilizing SMAD2 and SMAD3, and dominant negative or Fc soluble ectodomain versions produce profound muscle hypertrophy, which is why this receptor anchors so much cachexia and muscular dystrophy pharmacology. Null animals die perinatally with cardiac and situs defects, so conditional deletion is required for adult muscle and metabolic questions, while ectodomain humanization gives an honest platform for testing bimagrumab class antibodies and engineered ligand traps against the human binding interface. Acvr2a shares ligands and compensates substantially, so interpret single-locus data cautiously. Choose conditional null alleles for lineage requirement, kinase-dead knockins for signaling dissection, and humanized ectodomains for biologic engagement, always with a validated tissue-restricted Cre.
