Bmp10, ortholog of human BMP10, encodes a BMP9-related ligand whose expression in the embryonic ventricular trabeculae and the adult right atrium makes it the most anatomically restricted member of the family. Processed dimers, including circulating BMP9 and BMP10 heterodimers, bind the type I receptor ALK1 with BMPR2 or ACVR2A and the endothelial coreceptor endoglin to phosphorylate SMAD1, SMAD5, and SMAD8, enforcing vascular quiescence and driving cardiomyocyte proliferation through Nkx2-5 and Mef2c. Germline nulls die near midgestation with hypoplastic trabeculae and arrested cardiac growth, so cardiac and endothelial conditional floxed alleles are required for any postnatal vascular malformation or pulmonary hypertension study. Loss-of-function logic here connects directly to hereditary hemorrhagic telangiectasia, where ALK1 and endoglin lesions phenocopy ligand deprivation. Because BMP9 encoded by Gdf2 substitutes substantially in circulation, compound Gdf2 and Bmp10 genetics is necessary to unmask ALK1 pathway collapse, and ligand-trap or antibody testing benefits from epitope-matched humanization.

Bmp10 conditional knockout mice carry a floxed allele so you delete function only where Cre is active. The germline allele stays intact until you cross to a tissue specific Cre.
