Cflar Knockout mouse models

Cflar encodes cellular FLICE inhibitory protein, the mouse ortholog of human CFLAR, a death effector domain protein recruited to FADD within death-inducing signaling complexes where it heterodimerizes with procaspase-8. The long isoform retains a catalytically dead caspase-like domain that paradoxically stabilizes partial caspase-8 activity sufficient for RIPK1 and CYLD cleavage while blocking full processing and apoptosis, whereas short isoforms truncate after the death effector domains and inhibit outright; abundance is set by NF-kappaB dependent transcription and rapid ITCH and cullin-3 mediated turnover, making CFLAR a labile rheostat between survival, apoptosis, and necroptosis. Germline deletion is embryonic lethal around E10.5 with cardiac and vascular failure, phenocopying Casp8 and Fadd loss, and is rescued by concurrent Ripk3 or Mlkl deficiency. Conditional flox alleles with keratinocyte, intestinal epithelial, or myeloid Cre are therefore required, and isoform-selective knockins are the tool of choice for separating long from short isoform function.

A conventional Cflar knockout removes gene function in all cells that inherit the allele. It is a proven first pass for target validation when redundancy is low.
