F5 Knockout mouse models

Mouse F5 is the ortholog of human F5 and encodes factor V, the nonenzymatic cofactor that raises factor Xa catalytic efficiency toward prothrombin by several orders of magnitude. Synthesized with A1, A2, B, A3, C1, and C2 domains, factor V is activated by thrombin or factor Xa cleavage that releases the large inhibitory B domain, allowing heterodimeric factor Va to bind membrane phosphatidylserine through its C2 discoidin domain and assemble prothrombinase. Downregulation occurs by activated protein C cleavage at Arg306, Arg506, and Arg679, and the human Leiden variant substituting Arg506 with glutamine confers APC resistance and thrombophilia, a mutation class best modeled by homologous knockin rather than null. Complete deficiency is lethal in mouse from embryonic and perinatal hemorrhage, so hypomorphic or conditional alleles are required, while the East Texas splice variant illustrates additional TFPI-related regulatory function. Pair Leiden knockins with thrombosis challenge endpoints and conditional alleles with platelet versus hepatic source questions.

Whole body loss of F5 gives the clearest readout when the question is whether the gene is required at all. Follow up tissue work can still move to a conditional allele if lethality or compensation appears.
