Fgf8 Conditional Knockout mouse models

Fgf8, ortholog of human FGF8, encodes a secreted ligand with multiple N-terminal splice isoforms of differing potency that signals through FGFR1c, FGFR2c, FGFR3c, and FGFR4 and functions as the organizing signal at the midbrain-hindbrain isthmus, the limb apical ectodermal ridge, the pharyngeal ectoderm, and the anterior neural ridge, coupling to ERK output that sustains progenitor proliferation and positional identity. Its biology is famously dosage dependent, which is why the hypomorphic neo-insertion allelic series remains the reference tool for producing graded craniofacial, cardiac outflow, and cerebellar phenotypes that complete nulls, lethal at gastrulation, cannot deliver. Human heterozygous variants cause Kallmann syndrome and normosmic hypogonadotropic hypogonadism through GnRH neuron deficits, and holoprosencephaly-spectrum defects also occur. Conditional floxed alleles with En1-Cre, Msx2-Cre, or Nkx2-5-Cre are essential for tissue attribution, and Fgf17 and Fgf18 overlap at c-isoform receptors in isthmus and limb. Match hypomorph, conditional, or humanized allele to whether the endpoint is patterning gradient, organ morphology, or ligand pharmacology.

Tissue restricted knockout of Fgf8 keeps wild type function everywhere else. Breeding is often more robust, and the setup mirrors patient biology where mutations arise in a subset of cells.
