Kmt2c Conditional Knockout mouse models

Enhancer methylation studies require Kmt2c, the mouse ortholog of human KMT2C (MLL3), a SET domain monomethyltransferase that, with its close paralog Kmt2d, forms COMPASS-like complexes containing NCOA6, PTIP, PA1, and the demethylase UTX to deposit H3K4me1 and license distal regulatory elements for p53, estrogen receptor, and lineage transcription factor output. Truncating and inactivating missense mutations accumulate across breast carcinoma, bladder cancer, medulloblastoma, and myeloid neoplasms, frequently as 7q deletions in the hematopoietic setting, so the disease-relevant allele is loss of function, often at haploinsufficient dosage, rather than hyperactivity. Model design consequently forks between conditional nulls that expose enhancer decommissioning and differentiation defects, SET-dead knockins that separate catalytic from scaffolding contributions, and humanized alleles for variant classification. Redundancy is real and quantitative, since Kmt2d covers overlapping enhancer sets, so compound conditional genetics should be planned upfront, and endpoint choice, whether tumor latency with a cooperating driver or chromatin profiling, dictates the appropriate Cre.

Tissue restricted knockout of Kmt2c keeps wild type function everywhere else. Breeding is often more robust, and the setup mirrors patient biology where mutations arise in a subset of cells.
