Ldhb Conditional Knockout mouse models

Human LDHB has as its mouse ortholog Ldhb, encoding the H subunit of lactate dehydrogenase, which forms tetramers with a lower Km for lactate and is inhibited by high pyruvate, biasing flux toward lactate oxidation and NADH generation in heart, red muscle and other oxidative tissues where imported lactate serves as fuel within the cell to cell lactate shuttle. This directional preference makes the isoform central to lactate consumption rather than production, and its loss shifts tissues toward pyruvate reduction and lactate accumulation; LDHB has also been implicated in lysosomal acidification and autophagy and is silenced by promoter methylation in several tumors. A conditional null with Myh6-Cre or Ckmm-Cre tests reliance on lactate oxidation under workload, whereas humanized knockins support isoform-selective inhibitor programs where sparing LDHA is the goal. Ldha co-expression yields hybrid tetramers retaining bidirectional activity, so quantify isoenzyme patterns. Align allele with arteriovenous lactate difference, lactate tracing, stress echocardiography and autophagic flux endpoints.

Conditional deletion of Ldhb limits the genetic change to the lineage you choose. That helps in oncology, immunology, and metabolic work where systemic loss would muddy the read. After you confirm Cre specificity, crossing to Ldhb floxed stock yields usable cohorts.
