Mmp14 Conditional Knockout mouse models

Mmp14 corresponds to human MMP14 and encodes MT1-MMP, the membrane anchored metalloproteinase that concentrates proteolysis at the cell surface through a transmembrane segment and short cytoplasmic tail directing it into invadopodia and controlling internalization and recycling. It cleaves type I, II, and III collagen directly, sheds CD44 and syndecan, and serves as the physiologic activator of pro-MMP2 using TIMP2 as a bridging cofactor, making it the apex node of pericellular matrix turnover. Germline nulls are severely affected, with dwarfism, craniofacial and skeletal dysplasia, fibrosis, and death by weaning, mirroring human MMP14 lesions that cause multicentric osteolysis nodulosis and arthropathy, so viability alone dictates conditional design. Use floxed alleles with endothelial, fibroblast, chondrocyte, or tumor cell restricted Cre lines to separate developmental from disease contributions, catalytically dead knockins to isolate proteolysis from receptor shedding, and humanization when antibody selectivity against the human catalytic cleft is the endpoint.

Tissue restricted knockout of Mmp14 keeps wild type function everywhere else. Breeding is often more robust, and the setup mirrors patient biology where mutations arise in a subset of cells.
