It drives primitive streak formation, mesendoderm specification, node function, and left-right asymmetry through an autoregulatory asymmetric enhancer, and human variants cause heterotaxy with transposition of the great arteries. Nulls arrest before streak formation, so conditional and hypomorphic alleles, or deletion of the asymmetric enhancer alone, are the only informative routes to later requirements. Gdf1 is not a spare copy but an obligate heterodimer partner for long-range signaling, so compound Gdf1 genetics and precisely timed Cre induction determine whether results are interpretable.
