Smad2 Conditional Knockout mouse models

Smad2, ortholog of human SMAD2, encodes a receptor-regulated SMAD for the TGF beta, activin and Nodal branch, phosphorylated on its carboxy-terminal SSMS motif by the type I receptors ALK4, ALK5 and ALK7 and then partnered with SMAD4. Its distinguishing feature is exon 3, a thirty-residue insert in the MH1 domain that blocks direct DNA contact, so the full-length protein acts through partners such as FOXH1, whereas the naturally occurring exon 3 deleted isoform binds SMAD-binding elements much like SMAD3. Germline deletion is early embryonic lethal with failure of egg cylinder patterning and mesoderm formation, mandating conditional nulls for adult fibrosis, immune or tumor endpoints. Exon 3 deletion knockins and carboxy-terminal phospho-dead alleles separate DNA binding mode from receptor input. Because Smad3 shares receptor input and SMAD4 partnership while binding DNA directly, phenotypes attributed to Smad2 should be tested against compound Smad2 and Smad3 genetics.

A floxed Smad2 allele paired with Cre gives spatial control that whole body knockouts cannot offer. People use it to separate developmental roles from adult homeostasis, to model somatic mutations, and to match disease that begins in one organ.
