Exon 3 deletion knockins and carboxy-terminal phospho-dead alleles separate DNA binding mode from receptor input. Because Smad3 shares receptor input and SMAD4 partnership while binding DNA directly, phenotypes attributed to Smad2 should be tested against compound Smad2 and Smad3 genetics.
