Retromer cargo selection depends on Vps35, the mouse ortholog of human VPS35, encoding the alpha-solenoid scaffold that binds VPS26 and VPS29 to form the trimeric cargo recognition complex, which associates with sorting nexins and the WASH complex to retrieve transmembrane cargo, including the cation-independent mannose 6-phosphate receptor, GLUT1, sortilin, and DMT1, from endosomes to the trans-Golgi network and cell surface. The dominant D620N mutation causes late-onset autosomal dominant Parkinson disease PARK17 by impairing WASH complex binding, disrupting endosomal actin dynamics and cargo trafficking, and elevating LRRK2 kinase activity, placing retromer directly upstream of established parkinsonian signaling. Since constitutive Vps35 loss is embryonic lethal, useful designs are the D620N knockin at the endogenous locus for the dominant mechanism, conditional null for retromer collapse in dopaminergic neurons or microglia, and heterozygous null for dosage sensitivity. Match allele class to LRRK2 substrate phosphorylation, cargo mislocalization, or nigrostriatal integrity endpoints.

Whole body loss of Vps35 gives the clearest readout when the question is whether the gene is required at all. Follow up tissue work can still move to a conditional allele if lethality or compensation appears.
