Why Model Il4

Il4 is the mouse ortholog of human IL4 and encodes interleukin-4, a cytokine produced mainly by T helper 2 cells, mast cells, and basophils. IL-4 signals through receptors containing IL-4Rα and has been associated with promotion of Th2 differentiation, B-cell class switching to IgE, and type 2 immune responses.

IL-4, together with related type 2 cytokines, is implicated in allergic and atopic diseases, and components of the IL-4/IL-13 pathway are targeted therapeutically. Il4-deficient mice are viable and show impaired Th2 responses.

Model Applications

Il4 knockout mice have been used to study type 2 immunity and allergic inflammation. Humanized IL4 models, often with humanized IL-4 receptor components, have been used to evaluate therapeutics targeting the type 2 cytokine pathway in defined experimental settings.
