Why model C4b
Mouse C4b, despite its name, encodes the functionally dominant murine complement C4 and is the practical ortholog of human C4B. The secreted alpha, beta, and gamma chain protein is cleaved by C1s or MASP2 to release C4a and expose a thioester in C4b that binds hydroxyl or amino groups on target surfaces, then recruits C2 for cleavage into the C4b2a classical and lectin pathway C3 convertase, with regulation by C4BP, factor I, CR1, and CD46. Human C4B favors ester linkage to carbohydrate, and copy number variation across C4A and C4B, together with endogenous retroviral insertion length variants, drives schizophrenia and lupus association through complement-dependent synaptic pruning. Because murine C4a (Slp) is hemolytically inactive, C4b deletion effectively creates whole classical pathway C4 deficiency without compound alleles. Use the null for immune complex and pruning phenotypes, and humanized knockins when isotype-specific covalent chemistry or copy number dosage is the actual variable.
